Umbellulone (CAS 24545-81-1) is a prominent monoterpene ketone from California bay laurel used mainly as a tool compound in pharmacology and neuroscience to probe migraine/headache mechanisms and neurogenic inflammation via TRPA1 activation, CGRP release, and meningeal vasodilation; it enables screening of TRPA1 antagonists and modulators of the CGRP pathway in in vitro assays (neuronal calcium imaging, electrophysiology) and preclinical in vivo models, and is applied in respiratory toxicology and chemosensory research to model nasal irritation, cough reflex, and airway hypersensitivity to environmental irritants; analytically, it serves as a marker/reference standard in GC–MS for authentication and quantification of Umbellularia essential oil and for studies of volatile emissions; it is also used as a reference scaffold in SAR around TRPA1 agonism rather than as a therapeutic due to its irritant and headache-inducing properties, necessitating strict handling, exposure control, and compliance with hazardous chemical regulations.
